Introducing SysQuan: Affordable Single-Injection Absolute Quantitative Proteomics using Stable Isotope-Labeled Mouse

Talk by Christoph Borchers, Scientific Advisor at MRM Proteomics 

Large-scale proteomics faces challenges like batch effects, long LC run times, and high costs of SIL standards. Our innovative workflow SysQuan, using SIL mouse as an internal standard, enables absolute human proteome quantitation, simplifying workflows and reducing costs. Human and mouse liver homogenates and plasma were digested and analyzed in a one-to-one mix, resulting in over 6,900 human liver and 1,400 plasma proteins in untargeted DDA-PASEF using the Evosep One coupled to a Bruker timsTOF HT. MRM quantified up to 300 hepatic proteins within a single injection, demonstrating the high performance of the Evosep combined with the Agilent 6495D.

Sequencing antibodies from de novo to identify functional and neutralizing antibodies in human plasma after SARS-CoV-2 vaccination

Talk by Thierry Le Bihan, Principal Scientist at Rapid Novor 

While B-cell sequencing has dominated SARS-CoV antibody discovery (accounting for 90% of findings in the CoV-AbDab database), this approach has limitations. It often fails to connect B-cells with circulating IgG antibodies, which are the primary agents of humoral immunity. Most IgGs are produced by bone marrow plasma cells rather than circulating B-cells. Although combining proteomics with B-cell repertoire analysis can confirm the presence of specific antibodies in circulation, it cannot identify antibodies missed by B-cell sequencing. De novo pAb sequencing, despite its technical challenges, offers a promising method to better understand the complete humoral immune response.
