The Evosep Approach: A standardized workflow that delivers high sensitivity and robustness
The Evosep approach standardizes the front end of the LC-MS workflow. Rather than requiring analysts to optimize every element independently, the workflow combines consistent Evotip loading with preconfigured methods and standardized separation.
By standardizing sample introduction and separation, the workflow is designed so laboratories can focus on scientific outcomes rather than front-end variability and routine troubleshooting.
Evosep Eno is a low-flow liquid chromatography system that uses Evotips – disposable sample-loading tips that standardize sample preparation and injection to support robust, reproducible LC-MS analysis.
Standardized and optimized methods can support:
Consistent sample introduction
Reduced operator-to-operator variability
Alignment between laboratories and sites
Predictable operation across extended sample series
A foundation for customer-led development and validation
No Compromise
Bioanalytical laboratories have traditionally faced a choice between the robustness of analytical-flow LC-MS and the sensitivity benefits of low-flow separations.
Traditional nanoflow workflows offer higher sensitivity, but often come with the operational complexity that limited broader adoption in regulated, high-throughput environments.
By standardizing the low-flow workflow, Evosep helps laboratories move beyond the traditional sensitivity-versus-robustness trade-off, supporting the sensitive, reproducible measurements required for PK/PD bioanalysis with the robustness and throughput needed for routine operation.
Why laboratories are moving beyond analytical flow
Evaluate your workflow and find out how Evosep technology can benefit your current needs:
Analytical Flow LC-MS
- Robust Operation
- Lower sensitivity
- Higher sample requirements
- Limited performance for challenging peptides
Standardized Evosep Low Flow
- Robust Operation
- Higher sensitivity
- Lower sample requirements
- Improved detection of low-abundance analytes
Low-flow sensitivity. Intended for routine bioanalysis.
For years, low-flow LC-MS has been associated with complexity.
The standardized Evosep workflow is positioned to address the operational challenges that have limited the use of traditional nanoflow in regulated, high-throughput laboratories.
Robustness and uptime
In high-volume laboratories, front-end troubleshooting consumes analyst time and instrument availability. The standardized, disposable-trap approach is designed to address operational challenges such as carryover, clogging and re-equilibration.
Reproducibility and standardization
Consistent Evotip loading and preconfigured methods are intended to reduce operator-to-operator and site-to-site variability, supporting reliable quantitative performance across development, validation and routine operation.
Throughput and utilization
Robust sample throughput can help laboratories use available LC-MS capacity more effectively, manage study backlogs and respond to changing sample volumes.
Specificity for evolving modalities
Low-flow LC-MS can support quantitation of challenging low-abundance peptides and surrogate peptides where sensitivity is critical.

From established antibodies to novel therapeutic formats
Many emerging biologics require greater assay sensitivity than analytical-flow LC-MS can routinely provide. Standardized low-flow LC-MS offers a practical path to improved sensitivity while maintaining operational robustness.
Monoclonal antibodies
Hybrid LC-MS/MS workflows can quantify a surrogate peptide released from a biotherapeutic following enrichment and proteolytic digestion. The measured surrogate peptide represents the intact protein within the defined assay workflow.
Surrogate and therapeutic peptides
Sensitive, reproducible separation is relevant for surrogate peptides generated from large-molecule therapeutics and for challenging peptide analytes.
Antibody-drug conjugates
ADCs can require measurement of multiple analytes, such as total antibody, conjugated drug and free payload. This creates a potential role for specific and multiplexed LC-MS quantitation.
Bispecific antibodies and engineered formats
Novel formats may lack readily available off-the-shelf LBA reagents. LC-MS can offer an alternative or complementary route when reagent availability, specificity or multiplexing is a challenge.
Evosep: A robust, validation-ready platform
Evosep provides a standardized platform that supports development, validation, transfer, and routine operation while maintaining the sensitivity advantages of low-flow LC-MS.

Analytical Development and Application of a
Targeted LC-MS/MS Pharmacokinetics Assay for Chimeric Aducanumab
Quantifying therapeutic monoclonal antibodies in preclinical studies is critical for understanding pharmacokinetics and tissue exposure but remains challenging in complex matrices such as brain tissue.
To overcome limitations of ligand-binding assays, the authors developed a targeted LC-MS/MS assay using parallel reaction monitoring (PRM) to quantify chimeric aducanumab (chAdu) in mouse plasma and brain cortex. The assay was validated and applied in a multi-dose pharmacokinetic study of a transgenic Alzheimer’s disease mouse model.

Evosep Eno – high sensitivity and quantitative accuracy
for plasma proteome analysis at scale
This application note showcases the Evosep Eno as a standardized platform for sensitive and reproducible targeted LC-MS workflows.
Combining automated plasma sample preparation with scheduled MRM analysis, the workflow demonstrated robust performance across 200 injections, low-amol sensitivity, and accurate quantification across a broad dynamic range. The study highlights how standardized low-flow LC-MS can support high-throughput quantitative workflows where robustness, sensitivity, and reproducibility are critical.
